Illuminating the brain: cutting-edge imaging solutions for neuroscience discovery

Dr. Stefano Abbiati - Dr.ssa Beatrice D'Ulivo - Dr. Giuliano Cazzola | Miltenyi Biotec Italia

Modalità di partecipazione: seminario in presenza con possibilità di collegamento remoto.

Light-sheet microscopy is a leading technology for 3D imaging of biological tissues.

TheUltraMicroscope BlazeTM (Miltenyi Biotec) enables high-resolution imaging of whole cleared samples, preserving spatial and anatomical context. Integrated with 3D immunofluorescence (3D-IF) and tissue clearing protocols, it allows detailed analysis of cellular architecture in neuroscience, oncology, and immunology.

A recent study by Bastiancich et al. (J Exp Clin Cancer Res, 2024) used this technology to investigate the post-surgical glioblastoma (GBM) microenvironment in mouse models. The study revealed a dynamic immune response and transient blood-brain barrier permeability after tumor resection. 3D imaging enabled visualization of microglial and myeloid cell distribution and morphology during early recurrence, identifying potential therapeutic windows for local and systemic interventions.

A further step forward is the integration of 3D imaging with 2D hyper-multiplex analysis using MACSimaTM Imaging Cyclic Staining (MICS). The 3D–2D workflow reveals regions of interest within the 3D volume and enables in-depth 2D molecular profiling, by detecting hundreds of protein markers simultaneously.

This approach offers unprecedented spatial and functional insights into tissue organization.

The seminar will highlight this integrated platform and its role in translational research workflows, with a focus on oncology and neurology for identifying novel therapeutic strategies.
